全書分為上下兩部分，前半部介紹博物院的業務概況，包括院史中的肇始、播遷、復院、擴建、轉型及茁壯等方面之重要記事，以及各單位的職掌與工作現況；後半部依年代由專門人員撰述解說本院部份精美藏品，盡括銅器、玉器、宋及清代瓷器、宋元明代書畫、緙絲刺繡、多寶格、宋版書、清代奏摺及台灣輿圖等。
